Yarra Ranges Highway Patrol are investigating following a single vehicle crash in Millgrove last night.
Investigators have been told a white Toyota Tarago was travelling south-west on Dee Road when it is believed the driver veered off the road and struck a tree about 10.30pm.
The driver and front seat passengers, both men, were airlifted to hospital with life-threatening injuries.
Four other people, three men and a woman were taken to hospital by road with non-life-threatening injuries.
The exact cause of the crash is yet to be determined and the investigation remains ongoing.
